Supposing I don't use prefix delegation and my clients get their IP address by manual configuration. Is it mandatory to configure IPv6 NAT with the -s parameter?

I assume you mean the MASQUERADE line? That's quite the setup you've got there.
Do you mind elaborating a bit more into this setup of yours?

Namely:

  • Do/will the clients share a gateway and LAN segment/cidr/etc?
